Peptides for Wrinkles: Seven Trials, and the Number That Travels Is the One a Person Guessed

Of twenty randomized records joining peptides to wrinkles, ten are oral supplements and one is not a peptide. Seven test a topical peptide — and where a trial ran both an instrument and a human grader, the figure that reached the marketing was the grader's.

The question has been tested. Not often, not in many people, and — this is the part that goes unremarked — not usually with the instrument the trial owned.

The census, and what it is mostly made of

PubMed, searched 2026-09-18, title and abstract terms, publication type randomized controlled trial: 20 records join a peptide to wrinkles or photoaging.

Half the randomized literature on "peptides and wrinkles" is about swallowing something. The oral collagen field is larger, better funded and methodologically more conventional than topical peptide research, and it dominates any search that does not exclude it. This count differs from the nine our peptides for skin census published on 2026-09-06, and the difference is the query rather than the field: that search was built to isolate topical peptides, this one deliberately was not, so that the exclusions could be counted and shown. Running it both ways is what makes the ten oral records visible instead of silently folded into a headline number.

One record contains no peptide. A 2011 study of Achillea millefolium (yarrow) extract matches because its abstract explains that proopiomelanocortin is a precursor peptide. It is an ingredient paper by authors at BASF Beauty Care Solutions France, which supplies the extract. A raw count is not a census until the records are read, and this one would have added a peptide trial that does not exist.

Five of seven owned an instrument. That is better than this site expected going in, and better than the category's reputation. The problem is downstream of the measurement.

The trial that ran both, and published one

The Argireline study is the cleanest test case in the literature because it did the honest thing twice over: 60 participants, randomized 3:1 against placebo, twice daily for four weeks on periorbital wrinkles, with both a subjective evaluation (Daniell's classification and Seeman's standard) and an objective one (silicone replicas of the treated skin, read by a wrinkle-analysis apparatus).

Here is what each arm reported.

Subjective: "the total anti-wrinkle efficacy in the argireline group was 48.9 %, compared with 0 % in the placebo group."

Objective: "the parameters of roughness were all decreased in the argireline group (p < 0.01), while no decrease was obvious in the placebo group (p > 0.05)."

48.9% against 0% is the figure that travels. It is on retail pages, in ingredient explainers and in AI answers. It is a global assessment by human observers — a count of how many faces someone judged to have improved.

The instrumental arm, which produced a physical measurement of skin geometry, is reported with a p-value and no effect size at all. How much the roughness fell is not in the paper's abstract. The number that could be compared against another trial, another ingredient or a threshold of visibility is the one that was not given.

This is not a claim of misconduct — it is a reporting convention, and a common one. But it produces a literature in which the most-quoted peptide statistic in cosmetics is an opinion count, while the measurement sitting beside it is unquantified.

The largest trial cannot answer the question

196 women, eight weeks, randomized, published in the British Journal of Dermatology — the biggest study in this set by a factor of three, and one of the few with a clinically meaningful comparator.

It compared a cosmetic regimen containing 5% niacinamide, peptides and 0.3% retinyl propionate against 0.02% tretinoin, and found the cosmetic regimen improved wrinkle appearance significantly more than tretinoin at eight weeks, with comparable benefit at 24 weeks and better tolerance throughout. Expert grading and digital image analysis were both used. The authors are at The Procter & Gamble Company.

It is a good trial of a regimen and it says nothing about peptides. Three actives shared the bottles, one of them a retinoid ester and one a vitamin with a far larger evidence file of its own, and there was no peptide-only arm. A reader looking for the strongest peptide wrinkle evidence is often pointed here, and this is the trial that cannot supply it.

Size, and who ran them

Excluding the 196-person regimen study, the trials that tested a peptide against a vehicle or placebo enrolled 60, 30, 22, 22 and 20. The median headcount across all seven where one is stated is 26.

Company authorship is disclosed and easy to check: OneSkin Inc. for OS-01 — every author — Nuritas Ltd for the AI-discovered pea peptide, Oriflame R&D Ltd for the triple peptide complex, Procter & Gamble for the regimen study. The post-laser study excluded from the count is authored by L'Oréal Research and Innovation.

Two details from those papers deserve to be read as written. The OS-01 trial was split-face against an identical peptide-free control, which is the right design — and its own abstract notes that texture and radiance improved "on both sides of the face", the vehicle side included. The 2020 pea-peptide paper describes its human component as a "28-day proof of concept pilot study" and its abstract gives no participant count, which places it outside what can be weighed at all.

The 2026 epidermal growth factor trial is a useful boundary case: 20 women, 12 weeks, investigator-rated, and its finding on the endpoint this page is about was that "global wrinkle severity ratings changed minimally in both cohorts." It also compared two active serums with no vehicle arm, so a null result there is uninformative rather than negative.

What this means next to the rest of the category

Our peptides for skin page counted the whole category against retinoids and found the ratio. This page is about the instrument, and the finding is narrower and more specific: the peptide wrinkle literature is better measured than it is reported. Five of seven trials built a physical measurement and the field quotes the graded percentages, because those are the ones published as numbers.

One endpoint over, the vocabulary problem repeats in a sharper form: firming and tightening claims turn out to belong to two other literatures entirely, one of energy devices and one of oral supplements, with a topical residue of six studies.

What would change this page

A single adequately powered randomized trial of one peptide against its identical vehicle, wrinkle depth pre-registered as the primary endpoint, measured instrumentally, run independently of the ingredient's supplier, and reported with an effect size. One paper would do it. As of 2026-09-18 the indexed record does not contain one, and the trial that came closest on design published a p-value where its number should have been.

Do peptides reduce wrinkles?

Seven randomized trials have tested a topical peptide against a wrinkle or photoaging endpoint in the indexed literature as of 2026-09-18. Most report a change in the direction the sponsor hoped for, in groups of twenty to sixty people, usually over four to twelve weeks. The category's problem is not that the trials are negative; it is that they are small, mostly run by the companies selling the ingredient, and that the figures quoted from them are usually the ones a human grader produced rather than the ones an instrument did.

How is a wrinkle actually measured in a trial?

Three ways, and they are not equivalent. An instrument records surface geometry — a silicone replica of the skin read by an analyser, fringe projection or profilometry building a three-dimensional roughness figure, or digital image analysis of standardised photographs. An expert grader scores the face against a published scale. And participants answer a questionnaire. The first produces a physical quantity, the second a trained opinion, the third an untrained one.

Which peptide has the strongest wrinkle trial?

On design, the Argireline study is the most informative single trial: 60 participants, randomized against placebo, and it ran both a graded assessment and a silicone-replica measurement. On what it published, it is also the clearest illustration of the category's reporting problem, because only the graded arm was given a number. No peptide in this literature has a large, independent, instrumentally measured randomized trial.

Why do the oral collagen trials show up in a search about topical peptides?

Because collagen peptides are peptides, and ingested collagen supplement research is a much larger and better-funded field than topical peptide research. Ten of the twenty randomized records returned on 2026-09-18 test a supplement taken by mouth — collagen hydrolysates, an elastin peptide from bonito, porcine placenta peptides, a caviar extract. Any count of peptide wrinkle trials that does not separate the two is mostly counting the supplement literature.

Are the trials run by the companies selling the peptides?

Several are, and the affiliations are printed in the papers. The OS-01 trial's authors are employed by OneSkin Inc., which sells it; the AI-discovered peptide paper's authors are all at Nuritas Ltd; the triple peptide complex study's lead author is at Oriflame R&D; the 196-person regimen study is authored by Procter & Gamble. Company authorship does not make a result wrong and it is disclosed openly. It does mean the field has almost no independent replication.

What would settle it?

One adequately powered randomized trial of a single peptide against its identical vehicle, with wrinkle depth as the pre-registered primary endpoint measured instrumentally, run by people with no commercial interest in the result, and published with effect sizes rather than significance alone. Nothing in the indexed literature currently meets that description, and the trial that came closest in design chose not to publish the number its instrument produced.
